In the best case\, these practices are embedded in day-to-day work\, not separate from it. Depending on the setti ng\, these practices can address everything from selection and design of a research project to decisions about how\, where\, and what to publish\; an d to promulgating results in policy and public settings. I will give a bri ef overview of the landscape of research integrity. In that context I will then focus my remarks on responsible data handling\, from project concepti on to terminal storage and assuring access in perpetuity. I will describe current institutional and political views on the value of record-keeping t ools such as electronic notebooks and data management plans. I will also d iscuss researcher\, institutional\, and funder responsibilities in helping to assure that good practices regarding data handling are made clear and e asy to implement.\n\nPlease note that Hamburg-based staff will be prioriti sed (as this has been arranged by the Hamburg Science & Society Committee) . It is highly studied in bacteria an d yeast\, and employed by viruses infecting mammalian cells. Even so\, its role in metazoan gene expression is underappreciated. A key player in tran scription termination is the cleavage and polyadenylation (CPA) factor PCF 11\, which directly binds to the Pol II CTD and can dismantle elongating P ol II from DNA in vitro. However\, PCF11 has hardly been studied in the ma mmalian system. We therefore employed various transcriptomic methods to sh ed light on genome-wide actions of PCF11. Our results indicate that PCF11 enhances both CPA as well as transcription termination in human cells\, pa rtially independent of each other. Surprisingly\, we found that PCF11 acts selectively\, preferentially targeting closely spaced coding and non-codin g transcription units that require efficient termination to avoid transcri ptional interference. Notably\, we show that PCF11 is sub-stoichiometric t o the CPA complex and acts as a regulatory factor. Low levels of PCF11 are maintained by an auto-regulatory mechanism involving premature termination of its own transcript\, and are important for normal development. Both in human cell culture and during zebrafish development\, PCF11 selectively at tenuates also the expression of other genes by premature CPA and terminati on. Strikingly\, this PCF11-dependent attenuation targets preferentially t ranscriptional regulators\, with the same genes prematurely terminated by PCF11 in human and zebrafish. The phenomenon of negative gene regulation b y premature termination is well described in S. cerevisiae\, where it play s a physiological role in response to changing growth conditions.
